Dominic Thiem was the defending champion, but withdrew from the tournament due to an ongoing wrist injury sustained in June at the Mallorca Open.[1]

World No. 1 and top seed Novak Djokovic is the only former major champion in the main draw to advance beyond the first round. A victory at the event would see him become the second man in the Open Era, after Rod Laver in 1969, to complete the Calendar Year Grand Slam.[2]

Djokovic is also attempting to win a 21st major singles title to break a tie that he holds with his Big Three counterparts, Roger Federer and Rafael Nadal, and become the sole holder of the men's singles all-time record total of major titles.[2]

By defeating world No. 3 Stefanos Tsitsipas in the third round, Carlos Alcaraz became the youngest player to beat a top 3 ranked-player at the US Open men's singles since the inception of the ATP Rankings in 1973. Additionally, he became the youngest player to reach the quarterfinals of any Grand Slam men's singles tournament since Michael Chang at the 1989 French Open and the youngest to do so at the US Open in the Open Era.[3] Félix Auger-Aliassime became the first player born in the 2000s to reach a Grand Slam semifinal.[4]

Botic van de Zandschulp became the first Dutch man to reach a singles Major quarterfinal since the 2004 Wimbledon Championships, and the first qualifier to do so at the US Open since Gilles Müller in 2008.[5][6]

With Djokovic and Medvedev both advancing to the final, this is the first time since 2015 that the 1st and 2nd seeds are contesting the championship match. By reaching the final, Djokovic also tied Federer's record of 31 major men's singles finals.[7]

§ The player did not qualify for the tournament in 2019 or 2020 and is defending points from a 2020 ATP Challenger Tour tournament (Ostrava) instead.
