The game features the characters of recurring Contra hero Bill Rizer and the humanoid alien samurai Genbei Yagyu from Neo Contra, and involves the two heroes fighting off the Neo Salamander Army led by Colonel Salamander.[4]

Contra ReBirth features a return to the 16-bit style graphics in its gameplay (like the previous Contra 4 did before), as well as remixes of music and tunes from the different games in the Contra series.[4] The game also features two-player cooperative gameplay.[5]
