Ashley Graham

Ashley Graham (voiced by Carolyn Lawrence) is the 20-year-old daughter of the newly-elected President, taken captive by the Los Illuminados during her trip home from Massachusetts and held in a European (possibly Spanish) village. It is Leon's initial objective in the game to ascertain her whereabouts and retrieve her. After meeting with her, it is revealed that she has been implanted with a Plaga parasite as well, so that the cult could gain control of her before returning her to the U.S.. The rest of the game then becomes a quest for Leon and Ashley to find a way to remove the parasites inside their bodies before they fall under Saddler's control. When Ashley is with Leon, it is the player's duty to protect her and prevent her from being carried away or harmed by the Ganados or other enemy creatures. As with previous Resident Evil games that feature short gameplay portions in which the player takes control of a supporting character, the player takes control of Ashley in Chapter 3-4 of the game.

Ashley is often viewed as a video game sex icon (many believe Capcom intended for her to be viewed this way as apparent by her extra outfit and the "up-skirt" easter egg). It is revealed in the ending cut-scene that she has a crush on Leon and even makes a sexually suggestive comment towards him.

Ada Wong

Luis Sera (voiced by Rino Romano) is a 28-year-old Spaniard who befriends Leon early in the game and teams up with him to escape the village. He claims to have formerly worked as a police officer in Madrid. In reality, he's a native of the village and was employed by Saddler as a researcher to find ways to strengthen the Plaga parasites. He betrays Saddler and steals the sample of Las Plagas, but eventually Saddler catches up with him and impales him with a barbed tentacle. Before dying, he gives Leon medication to suppress the growth of the Plaga in his body and leaves behind all of his research notes throughout the castle and island, which provides hints on the behaviour of Las Plagas and how to destroy many of the enemy creatures introduced during the course of the game. The plaga-removing machine which Leon and Ashley use near the end of the game was invented by him.

The "Separate Ways" scenario and "Ada's Report" documentary added to the PS2 version of the game expands on Luis's role in the game, depicting him as in league with Ada Wong and the organization she works with (a connection that was only implied in the main game).

Ingrid Hunnigan

Ingrid Hunnigan (voiced by Salli Saffioti) is Leon's primary radio contact and his only link to headquarters. She provides the player with general gameplay tips and hints between chapters early in the game. Leon loses contact with her after he first enters Ramon Salazar's castle in which the communication line is hijacked by Los Illuminados. Salazar (and later Saddler) takes over the transmission to taunt Leon between missions. Leon does not regain contact with Hunnigan until after the ending credits, which consists of Leon flirting with Hunnigan and her turning down his advances.

Jack Krauser

Jack Krauser (voiced by Jim Ward) is a mercenary hired by Saddler and is the one responsible for Ashley's kidnapping. A former U.S. agent, he was once a close friend of Leon until he was presumed dead two years before the events of the game and disappeared. In reality, he infiltrated the cult in order to procure the sample of "Las Plagas" and is actually working for Albert Wesker. In addition to being a skilled knife fighter and archer, Krauser is also endowed with the ability to transform his left arm into a giant mutated sword-like claw. Leon fights him twice during the course of the game: first in an interactive cut-scene which depicts a knife fight between the two, and later in an in-game battle across a set of ruins on the island. At the end of this battle, Krauser is assumed dead. However, in the 'Separate Ways' mini-game he battles Ada in his defeated form, suggesting that this battle takes place after he battles Leon for the second time.

Krauser appears as the end boss in the "Assignment: Ada" mini-game. He is also a playable character in the "The Mercenaries" mini-game, in which his claw can be used as a special attack by the player. The "Separate Ways" scenario added to the PS2 version depicts a third battle with Krauser occurring during the main story, this time against Ada. In addition, the "Ada's Report" documentary expands on Krauser's role in the storyline.

Krauser bears a distinct resmemblance to a character from another well-known Capcom franchise, Final Fight/Street Fighter's Rolento. Despite mirroring each other in several respects (fighting style, red berets, use of knives and grenades), there is no apparent connection between the two characters.

Osmund Saddler

Osmund Saddler (voiced by Michael Gough) serves as the main antagonist of Resident Evil 4. He is the current leader of a religious cult known as the Los Illuminados, which seeks to control the world by overthrowing the U.S. Government and intends to use Ashley for this purpose. He has converted the villagers into his cult and now controls them due to the Plaga parasites in their bodies. It is believed by Ada's organization that his ceremonial rod allows him to retain control of the Ganados and the other creatures. His main underlings are Bitores Mendez and local castellan Salazar (whom he converted to his cult), in addition to Jack Krauser. The final battle between Leon and Saddler takes place in an island base located outside the village, in which Saddler transforms into a spider-like creature. His main weak spot is the eye on his mouth; primarily he can be killed with a specialized rocket launcher provided by Ada, or by a continuous assault on the eye (allowing the player to keep the specialized rocket during their next playthrough). If the player is playing Separate Ways, then Saddler's original body is fought in the cryogenic chamber where Ashley was held. Saddler moves at an inhumanly fast speed and can knock the player in multiple directions. He can absorb bullets, and shoot them out of his hand with the same force as if they were fired through the barrel of a gun. Most notably, he can stretch his arms across the room in sharp tentacles which can either impale Ada, or allow him to reach the catwalk on the second floor.

Bitores Mendez

Bitores Mendez (voiced by Jesse Corti) is the village chief whom Leon faces at the beginning of the game. Some of his distinguishing characteristics include his imposing stature, fake left eye and apparent resistance to firearms. He leads the hunt for Leon and Ashley early in the game, leaving behind notes of his strategies to trap the two. Bitores's normal human body is never fought one-on-one. Instead, his mutant form is fought. In this form, Mendez sprouts four tentacle-like spears from the flesh on his back, and his body is essentially torn in half as his spine extends upward,like a worm with many legs,and adding an extra ten feet to his size. After killing him, the player can take his fake eye, which is needed for the retinal scanner that unlocks the gates to Salazar's castle. There is also an extra cutscene with Mendez. When Leon first meets Mendez outside of his room, Mendez chokes Leon and soon lets him go and enters his room. You can then go back into the room where another cutscene occurs. Only this time Ada comes to Leon's assistance.

Ramon Salazar

Ramon Salazar (voiced by Rene Mujica) is the eighth castellan of the castle which serves as the setting of the second third of the game. Ramon's ancestor, the first Salazar castellan was the one responsible for sealing the Las Plagas parasites in his castle centuries ago. Despite his diminutive stature and wrinkled face, he claims to be only 20 years old. He is protected by two cloaked bodyguards known as Verdugos (Executioners), one of whom Leon fights in the game (Salazar's "right hand"). He taunts Leon through his exploration of the castle, setting him up against various traps and creatures, before he and his remaining Verdugo merge with a mutated plant to fight Leon. Salazar's overall design bears a clear resemblance to Napoleon Bonaparte.
