  • Are each of those bold alternative names created as plausible redirects to this article?
  • "34.6-kilometre (21.5 mi)-long route" vs "33.8 km (21.0 mi)" (in the infobox)
  • "1934–June 1, 1970" spaced en-dash here.
  • "in 1934. In 1961" bit repetitive.
  • "January 1, 1998 and " comma after 1998.
  • "39 began began in" so good it began twice.
  • "Maidstone, Rochester and Sandwich East townships" none of these significant enough for articles?
  • "the Detroit–Windsor tunnel.[20]" overlinked.
  • "#2" do you mean literally "#2" or just "number 2"? Avoid using # to mean number per MOS.
  • "E.C. Row Avenue " spaced E. C.
  • "of Highway 401,Highway" something odd going on here.
  • "the Ministry of Transportation of Ontario. The" overlinked.
  • Refs 3, 19 and 23 have spaced hyphen, should be spaced en-dash.

Alright... at this point I'm going to be smart and start taking notes on these little style fixes, which I should be applying through every article I take care of. Of note (let me know if I'm being too broad):
  • All bold terms should have redirects
  • Ensure length matches throughout the article (duh hehe... it happens, that's what peer review is for!)
  • Check for overlinking
  • No "#", that's for twits
  • Space between X. X. Blah for names (should it be a non-breaking space between the two initials?)
  • Almost nothing should have a hyphen; use an em-dash, and maybe once in a blue moon you'll be wrong
Anyways, that said, I've addressed all of your points in the article, except for the overlink of the Ministry of Transportation; the link in the opening sentence for the intersection list is a consistency thing in every highway article as far as I'm aware.
