- September 1 - Murder Mystery plays its first official show as a four-piece granite work table at Mercury Lounge.

- September 3 through September 4 - The 40th Annual MDA Telethon hosted by Jerry Lewis to raise money for the Muscular Dystrophy Association.
- September 4 & September 5 - September 4 is Labor Day in the U.S. Steve Irwin, Crocodile Hunter, died from a stingray wound. R&B superstar Beyoncé to release her highly anticipated sophomore studio album, B'Day worldwide and in some U.S. stores September 4, and fully in the U.S. on September 5.
- September 5 - 'The Beatrice Letters', a spin-off of Lemony Snicket's widely acclaimed saga 'A Series Of Unfortunate Events' is due for release.
- September 5 - Katie Couric debuts on the CBS Evening News.

- September 5 - Alan Mulally suceeds William Clay Ford, Jr. as President and Chief Executive Officer of Ford Motor Company.
- September 8 - Star Trek celebrates its 40th Anniversary.
- September 10 - The Simpsons' 18th Season premiere will air on FOX.
- September 10 - Michael S. & Bernadette Wisniewski celebrate their 40th wedding anniversary at age 57.
- September 11 - 5th Anniversary of 9/11.
- September 12 - Apple will be revealing the future for iPod, iMac, iTunes, and more. Apple will also be revealing "One more thing".
- September 12 - Emily Haines of Metric to release her debut solo album Knives Don't Have Your Back.
- September 12 - John Mayer to release his studio album Continuum. Justin Timberlake to release his second album FutureSex/LoveSounds .
- September 12 - Yo La Tengo to release their 10th full-length album I Am Not Afraid Of You And I Will Beat Your Ass.
- September 12 - The Mars Volta to release their 3rd full-length album Amputechture.
- September 12 - The Black Keys to release their 4th full-length album Magic Potion.
- September 12 - Papa Roach release 4th major label album The Paramour Sessions.

- September 14 - Nintendo of America will hold a six-hour press conference in New York City at the Nintendo World Store.
- September 15 - Nintendo of Europe will hold a press conference in London, England.
- September 15 - UPN is expected to completely shutdown operations of the network in advance of the merger with The WB and the launch of The CW the next week. It is unknown what, if anything UPN will air to wrap up the network's operations. The date of the shutdown may also change, as this is the latest UPN can operate before the merger.
- September 16 - Gem Bailey and Tom BomB (in a relationship) are going to meet for the first time.
- September 17 - The WB finalizes complete shutdown of the broadcast network. It plans to end programming with 5 hours of the most memorable programing and image campaigns from the network's 11 year history one day before it merges with UPN to form The CW.
- September 18 - The CW debuts.
- September 18 - Pokemon Mystery Dungeon comes out in the US.
- September 20 - The first season premiere on The CW airs, the cycle premiere of America's Next Top Model, marking the first new episode of any show on the new broadcast network.
- September 22 - WWE Friday Night SmackDown! debuts on The CW.
- September 23 - Kids' WB! debuts on The CW as well as new shows called Tom and Jerry Tales, Monster Allergy, Legion of Super Heroes and Shaggy & Scooby-Doo Get A Clue!. Mass demonstration scheduled against Tony Blair and the Labour Party in the UK, to take place outside the Labour Party conference in Manchester.

- Weird Al Yankovic releases his 12th album, Straight Outta Lynwood
- Per federal court injunction, the Bush Administration must by September 2006 either provide evidence for why the visa of Tariq Ramadan should be denied, or else must issue a visa for him.
- September 28 - The first fourth-generation Pokémon games, Pokémon Diamond and Pearl are scheduled to be released in Japan.
